چکیده

The existence of a trypsin-like enzyme (TLE) in acrosomes ofepididymal spermatozoa was confirmed and was further demonstrated to be present in acrosomes of ejaculated and capacitated spermatozoa. TLE rapidly removes the zona pellucida of the ovum. Extracts of acrosomes of ejaculated spermatozoa contain an inhibitor that is separated from the TLE by purification of the TLE. The inhibitor of TLE is also present in seminal plasma. This enzyme\p=m-\ enzyme inhibitor relationship appears analogous to the corona-removing enzyme-decapacitation factor relationship and part of capacitation very likely involves removal of the inhibitor from TLE and decapacitation factor from the corona-removing enzyme. TLE is inhibited by soybean trypsin inhibitor and less effectively by mercaptoethanol.

A collagen-binding protein on the surface of ejaculated rabbit spermatozoa

A rabbit antibody to mouse 3T3 cell fibronectin was used in conjunction with a fluorescein-tagged second antibody to detect fibronectin-like activity on the surface of rabbit spermatozoa. Only ejaculated sperm displayed an intense and highly localized fluorescence over the acrosomal region. Cauda epididymal sperm of the rabbit as well as several other species did not exhibit any reaction. The f...

Novelty of the pyruvate metabolic enzyme dihydrolipoamide dehydrogenase in spermatozoa: correlation of its localization, tyrosine phosphorylation, and activity during sperm capacitation.

Spermatozoa are cells distinctly different from other somatic cells of the body, capacitation being one of the unique phenomena manifested by this gamete. We have shown earlier that dihydrolipoamide dehydrogenase, a post-pyruvate metabolic enzyme, undergoes capacitation-dependent tyrosine phosphorylation, and the functioning of the enzyme is required for hyperactivation (enhanced motility) and ...

O-6: Progesterone-Induced Reorganization of NOX-2 Components in Membrane Rafts is Criticafor Sperm Functioning in Capra Hircus

Background: Reactive oxygen species (ROS) is generated by spermatozoa from human (Aitken and Clarkson, 1987; Alvarez et al., 1987), rat (Vernet et al., 2001), mouse (Fisher and Aitken, 1997), rabbit (Holland et al., 1982) and buffalo (Roy and Atreja, 2008). ROS initiates signaling pathways regulating key processes including capacitation (Aitken et al., 1998;Aitken et al., 1996;deLamirande et al...

Desmosterol, the main sterol in rabbit semen: distribution among semen subfractions and its role in the in vitro spermatozoa acrosome reaction and motility.

Sterols are essential components of the cell membrane lipid bilayer that include molecules such as cholesterol and desmosterol, which are significantly found in the spermatozoa of several animal species. However, the presence of desmosterol in rabbit semen has never been investigated. The aims of this study were to characterize the sterol composition of subfractions of ejaculated rabbit semen a...
